Activités de recherche

Mon domaine d'expertise concerne les filaires (nématodes Onchocercidae, parasites de vertébrés), et leur endosymbionte Wolbachia. Plusieurs espèces sont des agents de graves maladies tropicales, comme les filarioses lymphatiques, l'onchocercose («cécité de rivières») et la loase.


Une autre particularité intéressante des filaires est qu'elles peuvent héberger une bactérie endosymbionte Wolbachia, présente aussi chez les arthropodes. Cette bactérie est actuellement une cible thérapeutique car les filaires ne survivent pas sans elle.


Nous nous intéressons à cette association symbiotique tripartite: un microorganisme eucaryote – Onchocercidae–, une bactérie endosymbionte – Wolbachia – et un organisme évolué –un vertébré terrestre– et aux processus d'adaptation au niveau moléculaire et cellulaire de ces partenaires les uns aux autres. Plus spécifiquement nous étudions:


1 - les relations phylogénétiques entre filaires et bactéries


2 - les migrations et le développement des filaires et leurs interactions avec l'environnement cellulaire (granulocytes, macrophages) et tissulaire (lymphatique, sanguin et pulmonaire) de leur hôte vertébré


3 - les réponses immunitaires du vertébré spécifiques de la bactérie et celles spécifiques de la filaire, et leurs modifications par des agents de stress, tels que des processus de vaccination, adjuvants ou des traitements anti-filariens/anti-bactériens.


Nous avons développé un modèle murin de filariose (Litomosoides sigmodontis ) que nous utilisons pour les points 2 et 3.


Actuellement nous nous intéressons à l'histoire évolutive des filaires d'oiseaux  et la coévolution entre collemboles et Wolbachia (collab. C. d'Haesse, UMR7179),  et nous étudions l'inflammation pulmonaire chronique dans le modèle de filariose avec L. sigmodontis.
